I got it to work this afternoon, actually! ! It's BLASTING as I type this smile

Were you able to get BLAST to work? I have been trying to BLAST my genome in DNA Master for the past few days and it has not worked. I was wondering if this was specific to my computer or the program.

Is there still a problem at Pitt? I have been trying to run an AutoAnnotation on DNA Master and although it says AutoAnnotation Complete there is not updates to the initial page. I have tried on multiple computers and it is not working. I have tried to update DNA Master but it is saying that there is an FTP Failure.

Some of our students are investigating potential lysogens. We have 2 questions:
1. Do we need to use 7H10 plates for the patch assay lysogen only plate, or can we use L-Agar plates without top agar? (if we do need the 7H10 plates, why is that?)
2. Can use use a wire loop rather than disposable loops?

I am trying to merge multiple files of our annotated phage. In DNA Master I am getting the following error "Cannot perform this operation on a closed dataset". I downloaded the files again and repeated the merge function, but got the same error. I was able to successfully merge 4 other phages, so I know my DNA Master software is working. It is most likely an error within the dnam5 files for the one phages where I see this error. Does anyone have an tips?

Another problem which may or may not be related. For one of our phages "ShiaLaBeouf" DNA Master says it has 253 genes, but when we run Starterator we get error messages for chunks of genes are missing. My questions is - if Starterator is says it can't find a gene (but it does exist in DNA Master), what does that mean?

When we Starterate our Phage, in our report it tell us say Geralt is found in track 153. But then when we go "look" for that track, it does not exist (No colors found, just black lines). Not sure what is going on. If someone wants to try it out, it is for Phage: Geralt_Draft, Gene # 13.
