Hi Debbie, Well the old computer died while I was attempting to use it. I was able to copy the files from what I could get from my work computer and then I downloaded the slip-stream update and put that into the files containing the old version of DNAMaster and it seems to be working, fingers crossed for the moment. Thanks for the suggestions and help as always. I will let you know if it stops again. Matt

Hi, I have been having trouble downloading DNAMaster on my new laptop and when it finally downloaded I had to go through Microsoft Edge and not Google Chrome. It is now giving me 3 error messages seen in the attached file. I have not had a chance to autoannotate any files as of yet, but when I put in all of the parameters mentioned in the bioinformatics guide it would not allow me to open any files and continued to crash and restart. I was able to downloaded the program again, but have not made any modifications to the parameters for Blast or any other areas. It is able to open previously autoannotated genomes. 3.03Mb

mastropm	I am annotating a EK1 phage Stormbreaker and the gp9 has both a Arsenate reductase and NrdH-like glutaredoxin domain in the same region. I am not sure what call to make on this since there is more evidence for the NrdH-like glutaredoxin based on the additional conserved domain database than the Arsenate reductase. I know that the Arsenate reductase is a newer function. 259Kb

mastropm	Hi Nikki, Yes we used your protocol. I think our nuclease mix is not 100% correct. We are getting a lot of what is most likely RNA contamination. The one I posted had the most smearing and the DNA concentration was too low. The one that worked had the least smearing of all. 92Kb

mastropm	We have had the same issues with streaky samples. We changed procedures and were able to increase yield of our samples. We still had some smearing and sent ours anyway and only had one sample pass QC for sequencing. Edited 30 May, 2019 18:28 68Kb
