jayadasgupta Yes, Jaya. For every lysate we prepare, regardless if its right after purification or later in aplification, it is always important to know the concentration of phage in that lysate so that we can know how much of that lysate to use in subsequent experiments. A webbed plate should yield a high titer, but until we know what that titer is, we shouldnt use it for experiments where particular concentrations are required. For example, you wouldnt want to extract DNA from a lysate if the titer is not at or above 10^9 pfu/ml. Short answer is yes, for every lysate. |
