Hello all!

When I try to "apply" the suggested changes, I keep getting a message that "Queries Table is Corrupt, reload from DNA Master FTP site?"

What does this mean and what needs to be done? Screenshot is attached.

For 2025 Genomics Workshop, Lindsay Constantini (from NCCU), was able to get a DNA Master installation to work on Crossover. Attached are the ChatGTP steps and some troubleshooting she used to install via Crossover on her MacOS.
best,
debbie

I am trying to use this cheat-sheet but I am stuck at
"1. In CrossOver, with DNA_Master_2705 selected:
2. Click “Open C: Drive” in the right panel.
3. A Finder window opens at drive_c.
4. Go to:
drive_c → Program Files → DNA Master"

When I click on Program Files, there is no option for DNA Master

Hello,

Do we have to calculate the titer again after the webbed plate is prepared and the lysate is collected?

Thanks,
Jaya.

Hello everyone,
I have been getting broth cultures ready from agar plates (isolated colonies). I was wondering if it is ok to subculture the broth Arthrobacter for next day's experiments? Meaning, take 1ml of the overnight broth and add to 50ml fresh broth for next day's lab? Or is that a no-no? And that we should start from the plate every time?.

Thank you,
Jaya.

The recommendation is to minimize the number of passages, as each passage increases the genetic variation in the population and possibly phenotypes important for infection. For a reliable culture, we recommend always starting a culture from a colonies off a freshly streaked plate. In a pinch, you can subculture from an existing culture, but I would do this regularly.
Thank you, Vic.
